Human iPSC-derived glutamatergic neurons expressing Cas9 for rapid gene knockout generation CRISPR knockout (CRISPRko)-Ready ioGlutamatergic Neurons are opti‑ox deterministically programmed excitatory neurons that constitutively express Cas9 nuclease. The cells arrive ready for guide RNA (gRNA) delivery from day 1 post-thaw. Using our optimised lentivirus or lipid-based gRNA delivery protocols, users can perform gene knockouts, pooled or arrayed CRISPR screens and start measuring readouts within a few days. Using CRISPRko-Ready, CRISPRi-Ready and CRISPRa-Ready ioGlutamatergic Neurons eliminates the need to spend months engineering and characterising Cas9-stable iPSC lines and optimising differentiation protocols, significantly reducing experimental timelines. With these ready-to-use cells, reliable and reproducible experimental results can be achieved by simply introducing gRNAs targeting the gene of interest. The cells are a powerful tool for functional genomics, drug target identification and translational research.
